Ksantohumol (Xn)

Common hops (Humulus lupulu s L.) contain approximately 130 different substances, including bitter acids, polyphenols, essential oils, monosaccharides, amino acids, proteins, lipids, fatty acids, pectins, salts, lignins, and other compounds. The polyphenolic compounds present in hops at an average level of 3-5% are represented by flavonoids.

The dominant flavonoid in hops is xanthohumol, which constitutes approximately 90% of all prenylated flavonoids, i.e. the so-called chalcones, in this plant. Its content in hop cones is approx. 1% of dry matter. Xanthohumol is contained in female inflorescences.

The extraction of natural xanthohumol from hops is a long-term process requmng specialist knowledge. Based on the expertise of a Professor of biological sciences, BioActive­ Tech Ltd. has developed an extremely efficient original method for extraction of xanthohumol with high pharmaceutical purity (http://xanthohumol.com.pl/). How does xanthohumol work?

Scientific investigations have demonstrated that xanthohumol exhibits antioxidant, anti­ inflammatory, antiviral, antibacterial, and antifungal activity as well as a broad-spectrum antitumor activity via inhibition of the growth of cancer cells in all stages of the neoplastic process.

Natural flavonoid compounds are known for their beneficial effects on the organism. In particular, xanthohumol, which is a flavonoid contained in hop cones, has a broad spectrum of biological activity [1]. It exerts antimicrobial effects through inhibition of the growth of e.g. Trichophyton mentagrophytes and T. rubrum molds, Gram-positive Staphylococcus and Propionibacterium bacteria, and DNA and RNA viruses [2]. Xanthohumol also has antioxidant activity, as it neutralizes free radicals and prevents inflammatory processes. It exerts anti-inflammatory effects via inhibition of cyclooxygenase (COX), lipoxygenase (LOX), and nitric oxide synthase (iNOS) enzymes [3].

Why is xanthohumol effective in the treatment and prophylaxis of inflammatory diseases?

The broad spectrum of the anti-inflammatory activity of xanthohumol (Xn) indicates its potential in the treatment and prophylaxis of inflammatory diseases, which is of key importance in the case of COVID-19 [4]. Investigations conducted to date have shown that the SARS-CoV-2 virus attacks human cells by targeting the angiotensin-converting enzyme 2 (ACE2) membrane receptor. The binding of the viral “Spike” protein to the ACE2 receptor present in the cell membrane mediates viral fusion and replication in the cell [5]. ACE2 receptors have specifically been detected in nasal epithelial cells, alveolar epithelial cells, lungs, and the lumen of intestinal epithelial cells. Hence, the nose, throat, lungs, and intestines are potential sites of viral invasion and penetration [6]. The largest numbers of ACE2 receptors are present in cells of patients with the so-called civilization diseases. The protective effect of xanthohumol on human umbilical endothelial cells damaged by the activated ACE2 receptor has been confirmed by scientific research results. Administration of Xn has been shown to produce a significant increase in the viability of damaged cells from 53.9 to 74.9% [7]. It should also be emphasized that Xn has been proved effective in alleviation of clinical symptoms and inflammatory reactions in lung tissues of farm animals [8].

Kurkumina (Cur)

Another chalcone, 1.e. curcumm with its derivatives called curcuminoids, is an ingredient of XopXn+Curcumin. Curcumin is a natural polyphenolic compound obtained from Curcuma longa rhizomes.

Similar to xanthohumol, it exhibits a number of therapeutic properties, e.g. antioxidant, antibacterial, anti-proliferative, anti-inflammatory, neuroprotective, and cardioprotective activities.

Curcuminoids, i.e. the most active ingredients in turmeric, are polyphenols with structural similarity to curcumin and very strong antioxidant activity responsible for the health­ enhancing properties of turmeric. As shown by the latest research, they increase the bioavailability and activity of curcumin [9].

How do curcumin and curcuminoids induce high immunity in the organism? What activity do they have?

As shown in a scientific publication from 2020 [1OJ, administration of curcumin may be a potential option to support the treatment of patients with coronavirus disease. Some potential effects of curcumin have been reviewed, e.g. inhibition of viral entry into cells and encapsulation, inhibition of viral protease, and modulation of various cell signaling pathways. Curcumin has been shown to impede SARS-CoV-2 entry and replication in cells. The protective effect of curcumin against the cytokine cascade has also been reported [11].

Importantly, curcumin is a very safe substance of natural origin. In an appropriate composition, it can contribute substantially to maintenance of homeostasis in the organism and delay the development of age-related diseases [12].

antioxidant, anti-inflammatory, and antihypertensive effects similar to Xn. Previous studies revealed high curcumin-induced levels of AT2R and ACE2 expression in myocardial cells, which evidences the protective mechanism of curcumin via modulation of the inflammatory effects mediated by ATlR and AT2R angiotensin II receptors. Similar effects were reported in xanthohumol studies.

Why is xanthohumol combined with curcumin and curcuminoids highly effective?

The medicinal product XopXn+Curcumin fonns a protective molecular film on the oral and throat mucosa, which prevents absorption of pathogens into the human organism. It is responsible for protection against pathogen infections and elimination of pro­ inflammatory processes.

The administration of the medicinal product XopXn+Curcumin in patients with COVID-19 was associated with significant improvement in saturation (administration of the product during the course of the disease) and absence of a decline in saturation (administration of the product from the beginning of the disease).

Noteworthy, hop flavonoids, e.g. xanthohumol, curcuminoids, and curcumin, do not dissolve in water but form a poorly absorbable suspension.
